Relaxed pRYG DNA: A topo II Cleavage Substrate lacking superhelical turns
pRYG DNA is isolated in a supercoiled form and then relaxed with topo I to produce topoisomers of the plasmid. Applications include:
- Its use as a marker for topo II assays.
- To replenish Drug Kits.
- To examine activity to non-supercoiled templates.
- To test for DNA intercalating drugs.
- To analyze DNA supercoiling (nucleosome assembly, gyrase action, etc.).
