Topoisomerase enzymes are sensitive to DNA quality (for example, heavy metal contamination) and not all DNA preps will support activity. All of our DNA substrates are screened to support topoisomerase (and gyrase) activity. When you do screening assays for DNA cleavages, you must have DNA with minimal contamination of nicked forms. Fortunately, TopoGEN controls plasmid and kDNA stocks for nicked contaminants, so you do not have to worry about wasting expensive enzymes on inferior DNA substrate.

Kinetoplast DNA (kDNA)

kDNA is the ideal substrate for topoisomerase II assays because it is specific for type II reaction mechanisms.  Researchers can even assay for a type II enzyme in the presence of large excess of topoisomerase I.
